Previous FBI head James Comey has denied the charges to making false statements to congressional representatives and hindering a congressional proceeding.
The former director's attorney entered the answer acting as his counsel in a national courtroom in Alexandria, Virginia on Wednesday.
His attorney declared he would try to have the legal matter thrown out for various factors including that his the accused, a opponent of President Donald Trump, was being singled out.
Mr Comey was formally accused just days after Donald Trump pressured his top legal official to move forward against him.
At the proceedings on Wednesday, his attorney Patrick Fitzgerald told the magistrate they aimed to submit multiple petitions to throw out the case before a court case, claiming the legal action was retaliatory and that a US attorney was wrongfully designated to take over the legal matter.
Mr Comey's case was originally handled by the original attorney, who stepped down under pressure from Trump after his investigation into a different political adversary - NY AG Letitia James - was unable to bring criminal charges.
James Comey appeared in positive mood as he entered the courtroom on Wednesday, chatting with his legal team and making jokes. He was joined by his spouse, his wife and child Maureen, a US lawyer who the previous administration terminated.
Following the reading to the judge state his legal protections and the pair of allegations against him in legal proceedings on Wednesday, James Comey was inquired if he understood the charges.
"I do your honour. Appreciate it," he told the judge.
James Comey was the chief of the Federal Bureau of Investigation from 2013 to 2017 and was fired about several months into the former president's first administration as national leader. At the period, he was directing an investigation into foreign election meddling and whether there were any links between Russia and the election effort.
While in office, Mr Comey triggered a backlash from the Democratic party when he announced just shortly prior to the national vote that he was examining presidential nominee the candidate's use of a non-governmental email account. Charges against Hillary Clinton were never brought, resulting in disapproval from conservative politicians as well.
Prosecutors assert Mr Comey gave untrue statements to the legislature during a congressional hearing in recent years when he was being questioned about both the email probe as well as the Russia election probe. They say he misled the Senate by claiming he had not permitted someone at the agency to leak to news outlets data about the FBI investigations.
Government lawyers also allege the defendant of "wrongfully trying to sway, block and slow" the panel by providing untrue information to it.
In a video James Comey posted to his Instagram after he was accused, he stated he was innocent and alleged Trump of conducting himself like a "tyrant".
"My family and I have been aware for years that there are consequences to opposing the former president," he commented. "We will not live on our knees."
"I'm innocent," he added. "Thus initiate a court case."
The accusations against the former director surfaced after Donald Trump shared on digital channels demanding his chief prosecutor, Ms Bondi, charge the former director and additional individuals.
